Thermal Shift Assays (TSA), or Differential Scanning Fluorimetry (DSF), is a label-free screening method used to determine protein properties such as thermal stability and ligand binding affinity. Two specific TSA applications will be demonstrated:

  • High-throughput analysis of protein thermal stability using melting temperature algorithms
  • Using a Boltzmann fit strategy on melting curve dilution series to determine ligand binding affinities between one compound and several targets
