Affinity Selection‑Driven Hit Filtering with Shape Scoring

Affinity selection mass spectrometry (ASMS) is a high-throughput approach for identifying ligand-protein interactions, but robust hit filtering remains challenging due to chemical noise, adduct formation, and overlapping isotopic patterns. Accurate assessment of isotopic envelopes is therefore critical to reliably distinguish true binders from analytical artifacts in ASMS screening workflows.

This poster presents an approach combining isotopic envelope shape distance scoring with retention time and mass accuracy criteria to improve hit discrimination in complex ASMS datasets, as implemented in Genedata Expressionist® for advanced mass spectrometry data processing and quality control, and leveraged in Genedata Screener® for integrated hit review and triage. You will learn how these integrated scoring and filtering strategies improve confidence in hit prioritization in multiplexed ASMS experiments, reduce false positives from noise, cross hits, and coeluting species, and streamline hit triage and confirmation workflows.
